Mrs. Wilken has been employed at IMH for seven years, the past three and half as Clinical Supervisor for Iroquois Home Health. In her new position, she will assume the role vacated by Nancy Dickenson, who retired after 41 years of service at IMH. Mrs. Wilken is active in local civic organizations and her church. She and her husband, David, have two children, Coleman and Shelby. They reside in Thawville. Sherri Sorenson, RN, has been named Clinical Supervisor for Iroquois Home Health. She replaces Jennifer Wilken, who served in that role prior to her recent promotion. A Watseka native, Ms. Sorenson graduated from the Nursing Program at Kankakee Community College. PEPSINIZED WHEY FOR L. ACIDOPHILUS solids increased in that lower counts were obtained on MRSO agar than on MRS agar. These effects could be due to inhibitory metabolic products produced from higher concentration of whey components. Increasing concentrations of whey components also might have created an "imbalance" among nutrients in the medium. For instance, the media with higher whey solids might require different concentrations of yeast extract, thiotone, or Tween 80 to attain maximum growth of lactobacilli. It was reported 6, 18 ; that lactobacilli require oleic acid for optimum growth. Whereas the requirement for oleic acid can be provided by addition of Tween 80, different concentrations may have been necessary to support maximum growth of the culture in media containing higher concentrations of whey components. The type of neutralizer used to maintain pH at 6.0 during growth of the culture could influence maximum populations attained. The type of neutralizer can influence greatly growth of other lactic acid bacteria 8, 10 ; . The neutralizer in this study 20% Na2CO3 in 20% NH4OH ; would allow slow release of CO2 during growth of the culture at pH 6.0 in the fermentor. Longsworth and Maclnnes 9 ; reported that CO2 was essential for the growth of L. acidopbilus. It is possible that a different neutralizer would have been more effective in the media containing higher concentrations of whey. Although there is no information of the optimum amounts of various minerals and salts on the growth of L. acidopbilus, an alteration of amounts of various salts caused by increasing whey solids could have influenced the culture. Lower stability of concentrated cultures in liquid nitrogen prepared from cells grown in the 5.0% medium could be from damage to the cells during growth. Damage or injury to the cells also is indicated by larger differences i.e. bile resistance ; between MRS and MRSO agar counts for the culture grown in the 5.0% medium than for the ceils grown in the 2.5% medium. Many factors can affect survival of L. acidopbilus in concentrated cultures during frozen storage. Smittle et aI. 12 ; found that cells of L. bulgaricus grown in a medium containing Tween 80 survived storage in liquid nitrogen better than cells grown in the medium without Tween 80. Goldberg and Eschar 5 ; and Smittle et al. 13 ; later reported that addition of Tween.

On the contrary, C18: 1 concentration decreased in response to freezing in lactic streptococci Gilliland and Speck, 1974 ; , and to spray-drying in Lb. acidophilus Brennan et al., 1986 ; . A high cycC19: 0 concentration favored the cryotolerance of Lb. bulgaricus, Lactobacillus helveticus, and Lb. acidophilus Gomez Zavaglia et al., 2000 ; . The cycC19: 0 also increased in response to acid stress Beal et al., 2001 ; , osmotic stress Guillot et al., 2000 ; , ethanol stress Teixeira et al., 2002 ; , and high age of culture Drici-Cachon et al., 1996 ; . Different methods are presently proposed to improve the quality of lactic and probiotic starters: the use of effective cryoprotectants Fonseca et al., 2003 ; , the use of adequate freezing and storage conditions Foschino et al., 1996 ; , and the selection of more-resistant strains Monnet et al., 2003 ; . Therefore, this work aims to adapt Lb. acidophilus, by applying different temperature and pH conditions during fermentation, to improve its resistance to freezing and frozen storage. The analysis of the membrane fatty acid composition of adapted cells should improve our understanding of the cellular adaptive response. MATERIALS AND METHODS Bacterial Strain and Media Freeze-dried Lb. acidophilus RD758 Danisco, Dange-Saint-Romain, France ; was stored at -20C. It was thawed for 10 min at 37C before inoculation, which was carried out at 3 106 cfu mL and adefovir.
