It appears that the ester moiety of carbenicillin indanyl sodium facilitates binding in comparison with either carbenicillin or ampicillin. Since the indanyl ester function changes carbenicillin from a very polar compound to a more lipophilic penicillin, the hydrophobic character of indanyl penicillin may assist in its transport through the gram-positive cell wall to the penicillin-sensitive site, the transpeptidase enzyme 4, 10 ; . Such facilitated transport is reflected in the MIC values of 0.05 to 0.08 for carbenicillin indanyl sodium compared to 0.3 to 0.8 for carbenicillin with S. aureus Oxford. This interpretation is in complete agreement with results obtained by Biagi and co-workers using chromatographic techniques 2 ; . They found that the more lipophilic penicillins and cephalosporins have better activity against gram-positive bacteria. Both carbenicillin indanyl sodium and carbenicillin were considerably more active in vitro against penicillinase-producing isolates of S. aureus than was ampicillin. By Rob Rulon-Miller In honor of its fortieth anniversary the Antiquarian Booksellers Association of Japan ABAJ ; sponsored a series of events in January, which included a lecture given partially in Japanese ; by ABAA President John Crichton on "The Current State of the American Antiquarian Book Trade, " and culminated in an international book fair Friday and Saturday, January 28-29. The staging of an international fair in Tokyo is in itself an event. The last such fair was held when ABAJ hosted the ILAB Congress and Book Fair in 1990, although both Yushodo and Maruzen have invited Western dealers on several occasions to "exhibit" books in controlled environments. This was John Crichtons first trip to Tokyo but hes ready to go back in an instant. I mention this only because this is how I felt after my first trip there in 1990. Ive been back at least twenty times since and over the years have come to recognize that Tokyo truly is one of the great book cities of the world. However, since a measurement error by one step or so is considered to be within the `acceptable' range, we should always bear in mind that such errors are inevitably included in the raw data. If greater weight is placed on higher concentrations, as in the above method, a measurement error by one step would result in a difference of 1 if was mistaken for 2, whereas the same error would result in a difference of 4 if was mistaken for 8. Therefore, the number of dilution steps was used instead of the concentration of drugs to prevent any weighting bias, and the author's score was less affected by artificial error and or arbitrary operation. When the drug efficacy score is divided by the number of strains, this gives a mean dilution-step difference for the two drugs. Using this value as a logarithm on base 2 as 2 mean drug efficacy ratio can be expressed. This modification may allow comparison among several studies performed using different strains, or studies conducted at different facilities. In the Figure, the range of concentration of drugs A and B is set from 0.25 to 16 mg L. However, some agents such as new quinolones cannot achieve concentrations as high as 16 mg L in vivo. The calculation range of MICs should be set at a more practical level e.g. 0.252 mg L ; , and the results could then be compared as `partial scores'. In addition, the score can be easily recalculated for selected isolates; for example one can compare a score for all isolates with that calculated from only resistant strains. If the drug efficacy score is expressed as the sum of the positive and negative parts, as described above, this provides several benefits. Consider a case in which the drug efficacy score is very small, despite a large number of strains; suppose, for example, that 100 strains were used and drug A was scored as only ` 5' over drug B by a calculation of ` 7 5', derived from the low scores of both drugs A and B. In this case, almost all strains appear to be positioned on the diagonal of the MIC correlogram, suggesting that the two drugs have almost equal efficacy in vitro. If two drugs give such results with most strains, it is not necessary to measure the MICs of both drugs for physicians; either one is sufficient practical equivalence ; . When the efficacy of one drug is predominantly greater than the other, as in ` 205 5 200', only the more effective one should be stocked in hospital, if it is also clinically efficacious. The most difficult case might be that in which the drug efficacy score is low for the number of strains used, but both the positive and negative parts are large, e.g., ` 207 202 5'. One of the possible explanations for this situation is that the pattern of drug resistance differs greatly from strain to strain, i.e., resistance is highly specific to the chemical structure of the drug. Another is that other factors that may affect drug resistance are present, but are not well controlled, or that there may be a great error in measurement of MICs. In either case, we cannot simply.

MATERIALS AND METHODS Strains and culture conditions. Strains and plasmids used in this study are listed in Table 1. P. aeruginosa and E. coli used for genetic manipulation were cultured in Luria-Bertani LB ; broth or on LB plates with appropriate antibiotics 100 g of ampicillin and 30 g of chloramphenicol per ml for E. coli; 200 g of carbenicillin per ml for P. aeruginosa ; , at 37C.
