Despite pioneering radio broadcasts in the region, the radio medium unlike press was slow to assume an indigenous identity, and failed to sever ties with the BBC until the 1950's. The study also suggests the private radio stations have been successful in capturing a significant segment of the audience share, with the Sinhala language stations Sri FM owned by the Edirisinghe group and Sirasa owned by Maharaja Broadcasting recording a 22.62% and 18.67% share respectively in 2003. Meanwhile the SLBC's national service Swadeshiy Sevay, the regional transmissions Rjarata and Ruhuna, and the modernised replacement of the SLBC Velda Seyay commercial service Pvana, have recorded only a mere 16.09% which includes a 10.35% share enjoyed by Swadeshiy Sevay. Meanwhile the SLBC's Tamil service Thendral recorded 3.77%, while the Asian Broadcasting Corporation's Soorian and the Maharaja owned Shakthi Tamil stations both rated higher with Soorian capturing 4.25% of the audience share and Shakthi reaching a staggering 11.21%. However Sri Lanka's privately owned radio and television stations have limited transmission capabilities, broadcasting only on the FM band, while the SLBC by comparison broadcasts island-wide on medium wave, short wave and FM bands. Another restriction on privately owned commercial radio operation is that the licensing of radio frequencies remains in government control. Yet despite their broad coverage capacity and their 10 radio channels and five regional stations, the SLBC is losing ground in metropolitan areas where its channels compete directly with private interests. The purified [ 3 H]Gal 6SO 4 ; pi4G1CNACR 6SO 4 ; was hydrolyzed m O . 100C for 40 min. After the partial acid hydrolysis, the sample was dried m a vacuum desiccator and dissolved in 100 \x\ of saturated NaHCO3. For W-reacetylarion, 2 uJ of acetic anhydride was added twice to the sample at an interval of 5 min and then 6 u.1 of acetic anhydride was added. After 15 min at room temperature, the sample was passed through a 0.2 ml column of Dowex 50H + and washed with 1 ml water. The eluate and washing were combined, dried, and subjected to paper chromatography. After developing for 40 h, a 3H-labeled peak migrating to the position of [3H]Galfil UjlcNAcR 6SO4 ; the second peak from the paper origin ; was collected and purified with paper electrophoresis. In balancing positive and negative signals after HNP stimulation. A striking finding in the present study is the dramatic effect of HNP on epithelial cell adhesion. We believe that the increased cell adhesion caused by HNP is mediated largely by the up-regulation of the ICAM-1 protein. The adhesion molecule ICAM-1 belongs to the Ig superfamily and possesses costimulatory activity in lung epithelial cells [58]. The interaction of LFA-1 with its ligand ICAM-1 is well known to be an important step in the activation of lymphocytes [59 61]. ICAM-1 plays a dynamic role in the initiation of CD4 lymphocyte and APC interaction compared with other adhesion molecules [15, 26], and a similar mechanism may apply in the present coculture model with lung epithelial cells. It was not our focus to investigate the relationship between ICAM-1 and MHC-I, but it is of interest to point out that ICAM-1 is associated with MHC-I proteins [62], and enhanced engagement of ICAM-1 on target cells leads to recruitment of the MHC-I proteins to the contact area and enhances presentation of cognate peptide MHC-I complexes to lymphocytes [15]. A continuous monolayer typical of the epithelial cells appeared to lose the cohesive phenotype, exhibiting stellateshape morphology with long, filopodia-like extensions after HNP stimulation in the coculture with CD4 lymphocytes. The exact mechanisms remain to be elucidated, but it has been shown that an altered modulation of Ig superfamily proteins such as junctional adhesion molecules can produce dramatic changes in epithelial cell morphology [63], which are similar to our observations in the present study. Further investigation is thus required to examine the effects of HNP on the regulation of junctional adhesion molecules. NF- B is involved in regulating the expression of many molecules in immune response pathways including MHC molecules, adhesion molecules, and costimulatory molecules. In this study, we showed that stimulation with HNP for 2 h resulted in an up-regulation of nuclear p50 in CD4 lymphocytes. We speculated that the increase in p50 activation was a negative signal to modulate the overwhelming immune responses induced by HNP, as p50 homodimers bind to histone deacetylase1 HDAC1 ; , and the p50-HDAC1 complex can then bind to DNA and repress transcription [64]. It is interesting that no significant nuclear p65 activation was found at the time that p53 was up-regulated after the stimulation with HNP. It is possible that the peak signaling of p65 activation was missed at an early time-point, which was associated with a degradation of I- B. However, a previous study showed that degradation of I- B is largely dependent on the CD28 costimulatory signal [65]. The NF- B signaling pathway is known to be important for activation of ICAM-1 transcription. Here, we demonstrate that HNP stimulation did not result in p50 translocation but induced p53 activation in A549 cells. Other investigators have reported that NF- B inhibition does not prevent the effect of p53 on ICAM-1 expression after DNA damage, and induction of ICAM-1 is abolished after treatment with the specific p53 inhibitor pifithrin- and is abrogated in p53-deficient cell lines [66]. Taken together, these results suggest HNP can modulate nuclear transcriptional factors contributing to immune responses and lovenox.

Dr. Margaret Spitz presented an integrative epidemiologic approach to studying the carcinogenic spectrum from propensity to cigarette smoking, through predisposition and early diagnosis, to outcome, by applying the principles of epidemiology to the notable advances in molecular genetics. Interindividual variation in susceptibility to tobacco carcinogenesis may explain why only a fraction of smokers will develop lung cancer. Tobacco dependence exhibits characteristics of a complex genetic trait with gene-to-gene and gene-to-environment interactions and phenotypic genotypic heterogeneity. Two examples of genetic variants modulating smoking behavior were presented--the dopamine receptor and nicotine metabolic pathways. One cellular process that may explain variation in tumor susceptibility is DNA repair capacity, which is critical in maintaining genome integrity. Her laboratory applies a functional assay to measure DNA repair capacity and there is a statistically significant trend for increasing lung cancer risk with decreasing DNA repair capacity. Functional assays are labor-intensive and not amenable to large-scale, high-throughput application in population studies, and her goal is to identify sequence variations in DNA repair genes in the DNA repair capacity pathway to predict the repair phenotype, and also to predict outcome in patients treated with platinumbased chemotherapy. How well markers in surrogate tissues peripheral lymphocytes ; reflect genetic events in the target tissue is still unknown. There is a need to develop novel repair assays, build robust multivariate models, and perform genotype phenotype and surrogate target tissue correlations and marinol.
