Freshly isolated myeloma cells were cultured for 2 days with 4 M of ATO, 100ng ml of APO2 TRAIL, or both and apoptosis was scored after 24 and 48 hours. The results are depicted in Table 2. As was the case for myeloma cell lines, the combination of ATO and APO2 TRAIL was better than additive in the induction of apoptosis in freshly isolated myeloma cells obtained from all 6 patients tested. Thus, following 24 hour of treatment, a mean percent apoptosis of 68.6 + 8.4% + S.D. ; was obtained for the combination of APO2 TRAIL plus ATO, whereas a mean of 31.8% + 9% and 23% + 6.2% was obtained for ATO alone and APO2 TRAIL alone, respectively. Rapid induction of differentiation in only 48 h 16, 24 ; . HPV31positive epithelial cells have been shown to express the differentiation markers involucrin and transglutaminase after suspension in methylcellulose as well as to express transcripts from p742, although no capsid proteins have been detected in this system 38 ; . Therefore, suspension in methylcellulose is a convenient system for use in a quick analysis of the effect of differentiation on a variety of life cycle events, even though it cannot facilitate the final steps of the viral life cycle. In addition to the other transcription factors mentioned, glucocorticoids have been shown to affect viral early-gene expression and promoter activity by binding to glucocorticoid response elements GREs ; in the URR 12, 14, 19, ; . Bromberg-White and Meyers have previously shown that the URR of HPV31 is not inducible by the synthetic glucocorticoid dexamethasone dex ; in monolayer culture 10 ; . In the present study, we demonstrate that suspension of the HPV31b-positive 9E cell line in the semisolid medium methylcellulose results in a dex-inducible activation of a luciferase promoter construct driven by the URR of HPV31 that was not seen in monolayer culture. The three GREs we identified by sequence analysis and their ability to bind the glucocorticoid receptor GR ; in the URR of HPV31 do not appear to be responsible for this inducibility. However, loss of all three GREs resulted in an increase in the induction, suggesting that each of the three GREs acts as a repressor of glucocorticoid action. By 5 deletion analysis, we localized the element s ; responsible for the dex inducibility of the URR of HPV31 to nucleotides nt ; 7238 to 7557. Furthermore, replacement of wild-type sequences between nt 7883 and 7900 by linker scanning mutagenesis resulted in an increased dex inducibility. Taken together, these results show that differentiation has a profound effect on the action of dex on the URR of HPV31, suggesting that glucocorticoids may play an important role in the differentiation-dependent life cycle of HPV and micafungin.

Coimplantation of small fragments of human fetal thymus and fetal liver into immunodeficient SCID mice resulted in the formation of a unique structure Thy Liv ; . Thereafter, the SCID-hu mice showed reproducible and long-term reconstitution of human hematopoietic activity. For periods lasting 5-11 mo after transplantation, active T lymphopoiesis was observed inside the grafts and cells that were negative for T cell markers were found to have colony-forming units for granulocyte macrophage CFU-GM ; and erythroid burst-forming unit BFU-E ; activity in the methylcellulose colony assay. In addition, structures similar to normal human bone marrow were observed inside the Thy Liv grafts, consisting of blast cells, mature and immature forms of myelomonocytic cells, and megakaryocytes . These data indicate long-term maintenance, in vivo, of human progenitor cells for the T lymphoid, myelomonocytic, erythroid, and megakaryocytic lineages. The role of the implanted fetal liver fragments was analyzed using HLAmismatched Thy Liv implants. The HLA type of the liver donor was found on T cells and macrophages in the graft . In addition, cells grown in the methylcellulose colony assay and cells in a bone marrow-like structure, the "thymic isle, " expressed the HLA type of the liver donor. Thus, the Thy Liv implants provided a microenvironment in which to follow human hematopoietic progenitor cells for multiple lineages. The formation of the Thy Liv structures also results in a continuous source of human T cells in the peripheral circulation of the SCID-hu mouse. Though present for 5-11 mo, these cells did not engage in a xenograft graft-versus-host ; reaction . This animal model, the first in which multilineage human hematopoietic activity is maintained for long periods of time, should be useful for the analysis of human hematopoiesis in vivo and mifeprex.
